⑶ 校准的检查。 在灭菌或使用过程中,很可能会使校准发生偏移。对于状况良好的传感器,这种偏移不会超过0.2个单位。但一些研究人员仍建议在发酵罐灭菌以后进行校准或者再校准。已有适用于较大发酵罐的这种系统,可以完全无菌地取出传感器,再将其部分地插入校准缓冲液中进行校准。在实验室规模下,有必要将传感器完全移出,利用前述的化学处理方法来进行再消毒。在实验室中检查一个可疑校准的较好方法是对发酵液进行无菌取样,在发酵罐外测量其pH值尽快进行检测、读数。因为细胞在不断变化的条件下(例如在连续培养中氧和基质的消耗)进行连续代谢,如果培养基的缓冲性能较差,pH在几分钟内即可发生显著变化,从而无法正确检查传感器的校准。
